The transformation of chloropurines into iodopurines is successfully achieved using cold hydroiodic acid,[‌141‌] e.g. the synthesis of 9-benzyl-6-iodo-9H-purine (181) (Scheme 103).[‌483‌] A chlorine at C2 is resistant to exchange, and upon reaction of 2,6,8-trichloro-9H-purine with hydroiodic acid (57% aq HI, 0°C, 45h), 2-chloro-6,8-diiodo-9H-purine is formed in 90% yield.[‌297‌]
