A microwave-assisted stepwise three-component synthesis starting from 2-aminobenzonitrile 103, dimethylformamide dimethyl acetal, and anilines gives 4-(arylamino)quinazolines 106 (Scheme 28).[25] This reaction is believed to go through quinazolinimine intermediate 105, which undergoes Dimroth rearrangement to furnish 106. Various anilines are suitable partners, giving quinazoline products in excellent yields, particularly for those substituted with electron-donating groups. Anilines substituted with electron-withdrawing groups tend to give a sluggish reaction with lower yields. Furthermore, electron-deficient anilines deactivated by a nitro group only generate the uncyclized intermediates.
