α-Ketoglutaric acid (2-oxopentanedioic acid, 25) is an important biomolecule; it is an intermediate in the tricarboxylic acid cycle, and a symbiotic substance in nitrogen fixation and molecular oxidation. It plays an important role in the metabolism of microbial cells and is also a key precursor for the synthesis of various amino acids and proteins. α-Ketoglutaric acid (25) can be prepared conveniently by the condensation of diethyl succinate and diethyl oxalate followed by decarboxylation of the intermediate triester 24 (Scheme 13).[‌17‌]
