The first asymmetric Mannich reaction catalyzed by an amine concerned the direct reaction of a ketone, an aldehyde, and 4-methoxyaniline (p-anisidine) in the presence of 35 mol% of (S)-proline in dimethyl sulfoxide. The good yields and excellent enantioselectivities obtained are the origin of the success of organocatalyst-mediated Mannich reactions.[2]
