In this chapter, the scope and the most recent applications of an ancient albeit very valuable multicomponent reaction, the Passerini reaction (and related processes), have been reviewed. Attractively, very mild conditions are used for these reactions and this allows a broad tolerance of many different functional groups, which only seldom need to be protected. The reaction indeed allows a fast entry to complex structures in just one step, perfectly adhering to the concepts of step and atom economy, making this reaction attractive also for industrial processes.
