In summary, the reversible immobilization of DNA-containing synthesis intermediates on a solid support can be of great value for the preparation of DNA-encoded chemical libraries. While early examples involved reactions that were, in principle, water compatible, in order to obtain enhanced reaction yields and for reasons of practicality, the use of strong anion-exchange resins allows reactions to be performed under anhydrous conditions.[4–8] Therefore, reversible immobilization has the potential to greatly expand the scope of on-DNA reactions and, correspondingly, their chemical diversity.
