The use of photo-crosslinkers and other electrophilic covalent linkers has become an important approach in DEL selections. The major advantage of this approach is that it establishes a stable connection between the binders and the target protein, thereby widening the difference between binders and non-binders in the aspect of stability on the target proteins. Thus, DPAL and other crosslinking-based methods are expected to see wide applications in DEL selections, especially against complex biological targets.
