Diversified DNA-encoded libraries (DELs) are based on universal building blocks. C—H activation methodology is a unique tool to enhance the value of building blocks by increasing the number of accessible reaction handles through atypical use of C—H bonds as handles. In addition to conventional transition-metal-catalyzed protocols, researchers have pioneered innovative approaches, including photocatalytic and biocatalytic methods, for the synthesis of DELs. It is imperative, irrespective of the chosen methodology, to ensure the preservation of DNA tag integrity throughout the reaction. In the authorsʼ view, the combination of electrosynthesis and reversible adsorption to solid support (RASS), a reaction modality that has not yet been published, would be a potentially useful tool for constructing DNA-encoded libraries.
